چکیده
To construct tissue-engineered blood vessels (TEBVs) in vitro, it is necessary to transfer seed cells three-dimensional (3D) scaffolds for culture. However, what happens the behavior of after they are transferred scaffold unclear. Therefore, this study, a transcriptome analysis was used characterize differentially expressed genes (DEGs) vascular smooth muscle (VSMCs) before and 3D polyglycolic acid (PGA) understand changes functional gene expression early stage Transcriptome sequencing results showed that DEGs were mainly enriched cell proliferation cell-cell adhesion. The grown PGA (PGA-VSMCs) signal transduction. Furthermore, we found ERK1/2 significantly activated PGA-VSMCs inhibiting phosphorylation level ERK 1/2 increased elastin. In conclusion, material altered VSMCs affected elastin production. This study advances our understanding biomaterial-cell interactions provides valuable insights improving cultivation TEBVs.

Intercellular communication in cultured human vascular smooth muscle cells.
Intercellular communication through gap junction channels plays a fundamental role in regulating vascular myocyte tone. We investigated gap junction channel expression and activity in myocytes from the physiologically distinct vasculature of the human internal mammary artery (IMA, conduit vessel) and saphenous vein (SV, capacitance vessel). Northern and Western blots documented the presence of ...

The role of oxidized high-density lipoprotein (oxHDL) and the protective effects of adiponectin in terms of vascular calcification is not well established. This study was conducted to investigate the effects of oxHDL with regards to inflammation and vascular calcification and to determine the protective role of adiponectin in attenuating the detrimental effects of oxHDL. Cell viability, mineral...
متن کاملTanshinone IIA inhibits AGEs-induced proliferation and migration of cultured vascular smooth muscle cells by suppressing ERK1/2 MAPK signaling
Objective(s): Vascular smooth muscle cells (VSMCs) play a key role in the pathogenesis of diabetic vascular disease. Our current study sought to explore the effects of tanshinone IIA on the proliferation and migration of VSMCs induced by advanced glycation end products (AGEs). Materials and Methods: In this study, we examined the effects of tanshinone IIA by cell proliferation assay and cell mi...
متن کاملEndothelial Differentiation of Human Adipose-Derived Stem Cells on Polyglycolic Acid/Polylactic Acid Mesh
Adipose-derived stem cell (ADSC) is considered as a cell source potentially useful for angiogenesis in tissue engineering and regenerative medicine. This study investigated the growth and endothelial differentiation of human ADSCs on polyglycolic acid/polylactic acid (PGA/PLA) mesh compared to 2D plastic. Cell adhesion, viability, and distribution of hADSCs on PGA/PLA mesh were observed by CM-D...
متن کاملThe Effect of Adiponectin on Matrix Metalloproteinase-9 (MMP-9) in Vascular Smooth Muscle Cells
Background & Aims: Atherosclerosis is a major cause of morbidity and mortality. Adiponectin reducesthe risk of heart disease, and matrix metalloproteinase-9 (MMP-9) is involved in the formation and development of atherosclerotic plaque. The aim of this study was the investigation of the effect of adiponectin on MMP-9 gene expression. It seems this hormone can reduce the risk of atherosclerosis ...
